I bought my first lithium battery (powerflite). It says to check it with a 1 amp load on battery tester. I've been using a 300mah for my nicad batteries. I'm using the battery on a 80" plane that is using 4 Hitec 5945s and one standard servo for throttle.

Can I use my 300mah set at 6 volts and be OK?

We suggest a 1-amp load for testing the Lithium-ion batteries and utilizing 6.9 to 7.0 volts as the stop fly point, which is based on using this load. This should give you a reasonable safety margin and plenty of flying during a day. You should contact the manufacture of the pack and verify this number. The 1-amp load testing started several years ago as the battery packs were becoming larger and with more energy density. The early load testers were designed when we were all using 600mah and 800mah packs. With the higher load, problems with any battery pack will become more evident thus preventing a failure that could cause lose of a model.
