Default OS300 4stroke twin

Could I replace the existing prop lock nut assembly that requires a 13.5MM hole be drilled through the prop with the prop nut lock assembly from an OS 1.6 which has the same 3/8-24 thread as the 300 twin but stops at the aft face of the forward large washer and would not require drilling the prop out to any larger than 3/8????

Four-stroke engines, with their strong tendency to kick back should use locking prop nuts. Also, there may not be enough propshaft for your propeller, washer, nut, and spinner if you change nuts. Better to drill the prop to the correct side. If the prop doesn't have enough material after drilling, you need to get a different prop.
