turn the needle all the way in and then out about 2 1/2 turns , this should get you in the ball park,

follow the breakin procedures that came with the engine

As highlander said, set the high speed needle valve first......then you might have to set the low speed needle valve also (most TT Pro .46's need to have this adjusted. If it loads up when idling (too rich), it will hesitate and sputter when transitioning to full throttle, and may also stall when idling for a while. turn the low speed screw in 1/8 turn at a time until it's right (it is located in the barrel that the throttle arm is attached to.

A good start is to read the manual, but as the guys say, 21/2 turns out should get you in the ball park

A GOOD break-in

TT engines typically benefit from a HOT break-in somewhat contrary to what their manual says to do...this is all per the late George Aldrich...this man forgot more than most engine gurus/manufactuters ever knew!

You should run this engine HOT just out of the box, two minutes, with a 9x6 prop @ 16.5K. ...then...after a complete cool down...go to an 11x5 for 3-5 eight ounce tankfuls at full throttle just slightly leaner than the rich/lean break setting. Then, put it on a plane and watch it get better and better.

...to do otherwise will end up with different results.

...do what you gotta do.

It is a ABC engine. As long as you don't run it 4-stroke rich, ,you should be OK. Make sure you don't run it too lean, too.
