I think I plugged my ESC on, up-side-down, would that kill it?

Shouldn't bother it. The positive lead is in the center of the three wires, ground and signal leads on opposite sides so if it's plugged in the wrong way, it normally just won't work. Unless you purchased that rx more than ten years ago and from a Horizon dealer, it's most likely a Chinese clone.
