I've been into RC for many years but have never used a 3 line fuel tank. If I fuel through the 3rd fuel line will it also send fuel to the carb?. I want to use a fuel dot setup so I don't have to mount a filler valve in the cowl. Thanks.

keep the throttle closed when ya fuel up. It should be adjusted to cut the engine off at the transmitter trim lever. Thats all......

I use 3 line tank set-ups on all of my planes as follows. I use 2 clunks and 1 vent line. One clunk line goes to the carb. The other goes to the fuel dot. And the vent line goes to the muffler pressure outlet. Very simple and trouble free..........RS

If your carb is way below the neck of the tank, then you might get just a little bit going that way. Otherwise the answer is no. You can test this out with the tank outside of the plane and not hooked up to the engine. Just fill the tank with fuel via the third line to see if fuel comes out the feed line.
