if you place the piston and the cylinder do you have to do another break in??

yup, you need to do a break in on it. just like a new motor

is all you replaced was the piston and sleeve?
what about the conrod, wrist pin, bearings, shims, gaskets, etc?
how much fuel was through the engine before the new parts?

i dont think so but the bottom bushing on the conrod blew and yes i replaced that but from a used engine
