The manual that came with the 6EXHP transmitter indicates the ESC should be connected at port 3 on the FP-R127DF receiver.
...how ever the Tx i purchased was packaged with the R146iP receiver, and no info on this Rx.
The R146iP receiver has connector #6 marked "6/B Power supply"
Does this indicate I should be using channel 6 rather then 3 for the ESC connection?

No. If you are using an electric speed control that provides power to the receiver, you connect it to the throttle channel. Power will be provided to the receiver and other servos. The slot you asked about is used with the regular switch harness. If you use a sixth servo, then you would use a "Y" connector and have a servo on one side of the "Y" and the regular switch harness on the other. Of course, you could do that with any channel.
