Absolutely. With a single pole connector (like you use on brushless motors), connect the + of the 1st battery to the - of the 2nd. Then connect the plus and minus of the series connected batteries to your ESC. Just like connecting two batteries in a flashlight. You double the voltage.

I suggest that you separate them when charging so you can use your conventional lipo charger that doesn't charge 6 cells.
