Set them wherever the engine runs the best.
Seriously, I assume you're talking about initial setup for starting. Set the idle needle so the head is just about flush with the throttle arm. Open the main valve 3 turns, this will provide a nice rich mixture for break in. You will tune the needles later, but for initial starting that's fine. Or you could just go to the Super Tigre website and download the manual, which will tell you the idle needle is set just fine from the factory and not to touch it untill you get the top end set.

You'll have to give it 4 or 5 tankfuls before you can get the low idle settings right.
Ringed Tigre's take a while to run in and give their best. The secret is, don't rush the engine, keep it nice and rich at 1st and then screw the High Speed Needle in a couple of clicks each tankful. When you have it leaned out nicely at the high end, then start fiddling with the idle mixture to get your transition right.

Get it started then adjust the high end for WOT and rich, then adjust the low end to get a reliable idle and transistion. I can't tell you exactly where to start. If I had to guess, I would say 2 turns out.
